It won't do any damage and as long as it's the same architecture it should boot OK if the media is good. Another check you could try - if you have a Linux PC to hand is to install the memory stick and mount the partitions and see what they look like. Failing that, download an alternative image - use the netboot for instance and see if you can boot from that. I've never heard of Rufus (Rufus Wainwright's about as close as I get), only ever used dd and never had any problems with it.
